I am unfamiliar with microscope video cameras, but have shot photos through my Nikon Eclipse/ D800E / Stackshot setup, etc. and have experimented with attaching a microscope objective to the DSLR. Thanks Pau, My skills in microscopy I would say are nominal. I did, for example, experiment with the condenser aperture some and realized closing the aperture down would make things look sharper, increase DOF (just like squinting an eye, or using a smaller aperture in photography), but I didn't know...

OK, I finally have StackShot working on my Nikon Eclipse E200 microscope, which have infinity plan acromat objectives. Thank you, Rik. I was able to get it working. It is a forgiving setup. I don't have a drill press, so my hand drilling for the shaft hole wasn't as parallel to the wooden cylinder sides as it should be, thus a slight wobble to the stepper motor :( but it works. I will replace the wooden shaft holder...
